Human p53 Antibody Product Summary

Specificity

MCA1842 recognises human p53, specifically binding only to p53 phosphorylated at the Ser392 residue.

Ser392 phosphorylation of p53 has been shown to occur following DNA damage. Phosphorylation of p53 at this site may be mediated the casein kinase 2 (CK2), and also be other phosphorylation mechanisms.

In immunohistology this antibody generally stains 5-10% of cells labelled with antibodies specific for non-phosphorylated p53.

For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.

This product requires antigen retrieval using heat treatment prior to staining of paraffin sections.

Where this antibody has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. It is recommended that the user titrates the antibody for use in their own system using appropriate negative/positive controls.

Storage

Store at +4oC or at -20oC if preferred.

This product should be stored undiluted.

Storage in frost free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
